You can check if someone is reading your mail but this possible only in gmail account.Check "return receipt" in the options when you send your email, and you will get a message back when he reads it.

ReadNotify lets you know when email you've sent gets read, It gives Instant Notifications : you will get an instant alert on your cell-phone or pager, via ICQ, or by email, the moment your email gets read.

ReadNotify will provide the following in tracking reports:
    • Date and time opened
    • Location of recipient (per their ISP city /town)
    • Map of location (available on paid subscriptions)
    • Recipients IP address
    • Apparent email address of opening (if available)
    • Referrer details (ie; if accessed via web mail etc)
    • URL clicks
    • How long the email was read for
    • How many times your email was opened
    • If your email was forwarded, or opened on a different computer

It is completely depend on the mail server, it is use to transfer computer program or software from one computer to another. To receive acknowledgement, you need option "Read Receipt" once the other person actually opens the email you send. The server recognizes that receiver has opened your mail, and then immediately sends you an email saying, "SoAndSo has read your email."
