Go in to chrome and click on settings, then options and at the bottom
you can set chrome as your default browser.

Check for any anti-spyware software that may be blocking the change.

Try not to make Chrome your default browser until they fix all the
security and privacy problems with it.

Maybe that is a good thing since chrome is still in Beta format.
You have to Run Chrome as Administrator.
After that you can go on with echrada´s reply.
Are they plugging into the Vista-standard "Default Programs" association
control panels found on the Start menu? That'd be the place to go for
one-stop shopping for associations.
It's pretty easy for applications to plug into this. =)

Right click on the Chrome shortcut and select Properties
Go to the Compatibility tab
Check "Run as Administrator"
Run Chrome
Click the "Allow" selection in the UAC prompt
In Chrome, go to Tools / Option and select "Make Chrome my Default Browser"
You may want to remove the Run as Administrator option as you will get hit with the UAC prompt every time you open Chrome or a new tab.
