Yes offcource, you can use usb drive to boost speed of Vista by the readyboost feature of Vista.

Ya i have used this readyboost, actually when you'll implement this it won't make much difference, and you'll not notice much difference in the amount of RAM you having now. In short it will give you boost in paging or indexing the files , but it won't give boost in applications.
Based on what i have read Pen Drive cannot be transform into RAM due to it's hardware structure. It is simply working like Harddisk
