You can do it if you're willing to use Compressed Folders, has the advantage of being native to the system thus requiring no extra software.

"Password protecting" a partition never works.
The only thing that really does work, and is very efficient, is encryption. Try TrueCrypt. It's free and it works like a charm.
Setting a password is not as possible but the access to a particular drive partition can be blocked for other login users through security tab settings. This can also be done at folder and file level.
Or else there are other third party softwares available that can give password protection at file and folder level, but not at hard disk partition level, like 'Folder Lock'. WinRAR can also provide such password protection. But the data needs to be archived in RAR format for that.
You can get free downloads of such softwares if you search in search engines. There might be such softwares available which can provide password protection for hard drive partitions but I am unaware.
