SP 2 is cumulative - you do not need SP 1 loaded first. But same time if you are using Vista You definitely required to have SP1 installed on Vista before you install SP2.I know Unlike most other Microsoft Service Packs, Vista SP2 is not cumulative.

Listen friend, "SP2 is an update to Windows Server 2008 and Windows Vista that associated improvements reveals through automated feedback, as well as updates that have been included since SP1. By providing these solutions incorporated into a single service pack for both client and server, Microsoft gives a single high-quality update that reduces deployment and testing complexity for it's users.

I would tell it is cumulative as most Microsoft service packs have been. Not sure though, since I installed it already having SP1 installed. It will report when you run pack 2 and skip one. So just go for it. It wont go through if it has to have 1 first. Its a update, it specifically looks that for itself.
