You are running with two of the options...

1- Since Exchange 2010 not able to support the Exchange 2000 so you may require to go for the upgrade Exchange to at least 2003 SP2, then you are able to perform the remote mailbox replacement to Exchange 2010 installation in different organization.

2- You are able to move for two step procedure , export the mailbox contents from Exchange 2000 organization and then after import the same to Exchange 2010 organization but you may require to be with the Outlook 2010 x64 bit version on workstation with the Exchange 2010 Management utilities installed to import the required mailboxes.

There can be some alternatives like you can go for the up-gradation of your specified messaging system to Exchange Server 2007 and then after jump on to the mailboxes to Exchange Server 2010.

You are able to make the transition to Exchange 2007 from the below suggested already implemented messaging systems:

    1. Exchange 2003 messaging system
    2. Exchange 2000 messaging system
    3. Collaborated Exchange 2003 and Exchange 2000 messaging system

Re: Migrate Exchange 2000 Data To Exchange 2010

When any pre-Exchange 2000 servers which may exist within your Exchange organization should be decommissioned before you are able to move to the native mode, it suggests which any Exchange 5.5 Servers in your organization should be completely removed before you are able to go with this steps. 'So does this suggests that you are unable to do a transition directly from Exchange 5.5 to Exchange 2007 in the similar Active Directory Forest?

Transition is the situation in that you upgrade an already Exchange organization to the Microsoft Exchange Server 2007. To go for doing the transition, you have to first go for the replacing the existing data from the existing Exchange servers with the new one Exchange 2010 servers. For instance , when you are doing a upgrade from the Exchange 2007 organization with an Exchange 2010 organization, the you need to do a transition.

It also based on the size means how small of an organization, and the capacity of the mailboxes in this case, you have to just go for the EXMERGE tools needed to do the export the components of the Exchange 2000 mailboxes, and then after import the evaluated PST file within Exchange 2010 with the help of the Powershell Import-Mailbox command.

You can use Lucid8's DigiScope to open an Offline Copy of the Exchange 2000 EDB and then migrate the data directly to Exchange 2010
