First, check out whether your internet connection is working fine or not. If you can connect to other website and unable to login your iTunes account, may because of the pop up blocker programs. These setting can affect the iTunes connectivity to the iTunes Store. By disabling the pop-up blocker will clear out this problem.
To disable pop-up blocker in Safari for Mac. Go to Safari and then Preferences. Select Security.Uncheck the box of pop-up windows
To disable pop-up blocker in windows.Go to Tools menu in Internet Explorer. Select Options and then Privacy tab. Uncheck the option of Block pop-ups.

There can be many behind your iTunes connection problem. The most important thing is that your will need to update your iTune application to the current date. You have to take care that you are having the lastest updated.
Update your iTunes software and try to login again.
Even after that if you are not able to login, then you have to remove the Genius feature of your iTunes software.
To remove the Genius feature run the Finder and go to the /username/Music/iTunes/ directory. Delete iTunes Library Genius.itdb file. Restart your computer and launch your application again.

In windows the computer uses the DNS setting to connect to internet. Because of this setting your iTunes connection is failed. To troubleshoot your problem you have reset the DNS setting of windows.
1). Steps to Reset your DNS setting in Windows XP:Go to Start Menu > Run > type cmd > type ipconfig /flushdns and press enter
2). Steps to Reset your DNS setting in Windows Vista:Go to Start Menu > All Programs > Accessories > Right click on command prompt > select Run as Administrator > type ipconfig /flushdns and press enter. Restart your computer. Now you can login your iTunes account.
