The correct IIS and NTFS permissions are documented in the WSUS Operations
Guide, and generally they are correct upon initial installation, so the
first step here should be to undo whatever you've changed. Of course, if
you've modified IIS permissions prior to installing WSUS, then they may not
be set correctly after the installation completes.

The second step, rather than arbitrarily changing permissions, is to
identify the actual *cause* of the error, and then apply the correct
remediation. In most cases, the cause of this particular error is that the
WSUS3SP1 installer failed to create the /selfupdate virtual directory in the
"Default Web Site" when the alternate "WSUS Administration" website is also
being used.
