Click on Start, go to Programs, go to Microsoft Exchange, and select System Manager. Expand all Servers, expand Server Name, and then expand Protocols. Expand SMTP, right-click the SMTP virtual server (for example, the default SMTP virtual server), and then put into Pause.
To resume the SMTP virtual server, right click on the SMTP virtual server (for example, the default SMTP virtual server), and then click Pause to erase the mark, which should show a break next time. If your queues continue to fill it is necessary to move the following patch http://support.microsoft.com/?id=835734

Follow this to delete selected messages from queues. If you delete a message from a queue, the message is permanently deleted from the mail system.

    1. Click Start, point to All Programs, point to Microsoft Exchange and then click System Manager.
    2. Double-click Servers, double-click the server you want, then click Queues.

Note If you have administrative groups defined, double-click Administrative Groups, double-click Administrative_Group_Name (where Administrative_Group_Name is the group that contains the queue you wish), double-click Servers, double - click the server you want and then click Queues.
    3. Right-click on the queue that you want to edit, then click Freeze to freeze the queue.
    4. Click Find Messages, and click Search to search for messages in the queue.
    5. Right-click on the message you want to delete, then click one of the following:

      • Click Delete (no NDR) to delete messages from the queue without sending a non-delivery report (NDR) to senders.
      • Click Remove (with NDR) to delete messages from the queue and send an NDR to the senders of messages.

Note To delete multiple messages that are at the other side, click the first message, hold down the SHIFT key and click the last message, right-click on the selected messages, and then click Delete (no NDR) or delete (with NDR). To delete several messages that are not on the other side, click the first message, hold down the CTRL key and click on other messages right-click on the selected messages and then click Delete (no NDR) or remove (with NDR).
    6. When you are prompted to delete messages in the queue, click Yes.

Re: How to Clear Queues Exchange 2007

use the Queue Viewer or the Exchange Management Shell to remove one or more messages in a queue on a computer server where the role of transport Hub or Edge Microsoft Exchange Server 2007 is installed. A message sent to multiple recipients can be found in several queues. To delete a message from several queues at once, you must use a filter. You can choose to send or not a non-delivery report (NDR) when you delete messages from a queue. You can not delete a message queue for submission.

Using the Queue Viewer to delete a message queue

    1. Open Exchange Management Console.
    2. In the console tree, click Toolbox.
    3. In the Results pane, click Queue Viewer Exchange.
    4. In the Actions pane, click Open the tool.
    5. In Queue Viewer, click the Messages tab. The list of all messages on the server you are connected is displayed. To adjust the action on a single queue, click the Queues tab, double-click the name of the queue, then click the Server \ Queue which 'poster.
    6. Select one or several messages from the list, right click and select Delete messages (with non-delivery report) or Delete messages (without non-delivery report). A dialog box confirming the selected action and asking Do you want to continue? Appears. Click Yes.
    7. To delete all messages from a queue, click the Queues tab. Select a queue, right click and select Delete messages (with non-delivery report) or Delete messages (without non-delivery report). A dialog box confirming the selected action and asking Do you want to continue? Appears. Click Yes.
