    PCI slot issue/not detecting card

I installed a new PCI USB controller card into the only PCI slot on the mobo, and it worked for about a week. It is plug-n-play so when I installed it, it installed the driver and worked fine. A few days later I went to use it again, and nothing. Looked in device manager, and it had disappeared. So I thought the card had gone bad. Contacted the company that made the card and they sent out a new one. Installed it and no joy. It wasn't detected by the mobo at all. I put the card in my other machine and it works fine. So this leads me to believe that something is wrong with the mobo/PCI slot. I have updated the BIOS, but that changed nothing.

This is a custom build that I built as an end of semester project for my hardware class. Here's the build list.

    *Windows 7 Professional 64 bit
*Sentey Extreme Division GS-6050 Halcon Black ATX Mid Tower
    *ASUS Sabertooth 990FX motherboard
    *AMD FX-8120 Zambezi 3.1GHz Eight-Core Desktop Processor
    *CORSAIR Vengeance 16GB (4 x 4GB) 240-Pin DDR3
    *SAPPHIRE Radeon HD 6570 2GB
    *Seagate Barracuda 7200.12 500GB
*COOLER MASTER GX Series 750W ATX12V v2.31 Active PFC Power Supply
*Logitech X-530 70 watts RMS 5.1 Speaker System
*LG Black BD-ROM Super Multi Blue with 3D Playback
    *LG 22X Super-Multi DVD Burner
    *LG Flatron 22' LCD montior
*Logitech MK550 Black Wireless Ergonomic Wave Combo

This is the PCI controller card that I installed.
    *Koutech USB 2.0 PCI (2 x Ext+2 x Int Header-pin) Model IO-PU222

I feel that it's the mobo too, but I was just really hoping that wasn't the case. I didn't want to have to deal with getting a replacement, but I guess I'll do what I've got to do. If it does have to be replaced, how will that affect the OS being attached to the old mobo? Will I be able to install the new mobo and all is good as far as the OS is concerned? I have a feeling I will having to be calling Microsoft to make it work.

Any help, suggestion, ideas will be appreciated. TIA!

From what you have explained it seems that the PCI slot on the motherboard has turned faulty and you will have to get it repaired or replace the motherboard.I don't think that you will have any issue with the new motherboard, if you want then you can take a back up to be at the safer side and make a fresh installation of the os later after getting new motherboard.

Yeah, I'm thinking it's the mobo too. I was just hoping it wasn't. I've contacted Asus and they want me to clear the CMOS and see it that helps. If not, then I'll have to send it in for a replacement. I really don't like haven't my main machine down for that long, but I guess there's nothing I can really do about that. I'll post update when I have anything new. Thanks for your help.
