The difference which you like to know for the speed between the USB 2.0 and the Esata,so i would like inform you that you would go with Esata, because esata provides six times speed of the USB 2.0 drive, the transfer rate of massive files is great,if the file size are in mb then the speed wont make any difference.

As compared to the usb 2.0, the External SATA (eSATA) hard drives provide additional speed for high bandwidth applications,eSATA cables are narrow and can be up to 6.56 feet (2 meters) in length, whereas parallel cables are much wider and limited to a length of 18 inches (45.7 cm),eSATA requires its own power connector.

The USB 2.0 is reliable, highly compatible, and easy-to-use, USB 2.0 is limited to 480 Mb/s, which translates into 30-35 MB/s maximum bandwidth for typical storage applications,This is certainly more than enough for most device types and for casual storage use
