AD 2003 to AD 2008 upgrade

I have recently built a test lab that has our 2 x windows 2003 DC's in it. I want to have a look at Windows 2008 Ad so I was wondering is it best for me to upgrade one of the test DC's to Windows 2008 or create a 3rd Windows 2008 member server and promote it? Thanks for any information.

I will always try to install an additional DC when new Operating System versions are used, even if inplace upgrades are fully supported from Microsoft. You can check the below website for more information.

Are you working with an elevated command prompt? You need to also ensure that you can contact the Infrastructure master as this is needed for the /rodcprep command. You can start the command prompt with Run as and try again. The "netdom query fsmo" would give you the IM listed with the DC having that FSMO, but as you said you already see in AD UC, it should be fine.
